Abstract publicado na página 172 do livro "7º Encontro Nacional de Cromatografia", do Departamento de Química e Bioquímica da Faculdade de Ciências da Universidade do Porto; ISBN: 978-989-97667-0-9
Structural and metabolic alterations in erythrocytes play an important role in the
pathophysiology of Chronic Obstructive Pulmonary Disease (COPD). Whether these
dysfunctions are related to the modulation of erythrocyte membrane proteins in
patients diagnosed with COPD remains to be determined. Herein, a comparative proteomic
profiling of the erythrocyte membrane fraction isolated from peripheral blood of smokers
diagnosed with COPD and smokers with no COPD was performed using differential 16O/18O
stable isotope labeling. A total of 219 proteins were quantified as being significantly
differentially expressed within the erythrocyte membrane proteomes of smokers with COPD
and healthy smokers. Functional pathway analysis showed that the most enriched
biofunctions were related to cell-to-cell signaling and interaction, hematologic...
The recent implementation of the Revised Bathing Water Directive in the European Union
has highlighted the need for development of effective methods to differentiate between
sources of faecal contamination. It had previously been shown that amplification of 16S rRNA genes of host-specific cacteriodales species using the HF183F and CF128F primers could be used as markers for human and bovine faecal contamination in the United States. This paper determined the sensitivity and specificity of these markers in four Atlantic Rim countries (France, Ireland, Portugal and the United Kingdom) to evaluate their usefulness in determining the origin of faecal contamination. It was shown that the HF183F marker displayed high sensitivity (80–100%) and specificity (91–100%), and is reliable as an indication of human faecal contamination. The CF128F ...
21 clinical isolates of Candida spp were examined by FT-IR in order to compare identification using FT-IR with biochemical identification (bioMerieux®). FT-IR revealed itself as a fast and effective method for identification of the strains tested as all were placed into the appropriate species, including one which was misidentified by the
biochemical method.
During the first two weeks of August 2003, Portugal was affected by a severe heat wave.
Following the identification in Portugal of the influence of heat waves on mortality in 1981 and 1991 (estimated excess of about 1900 and 1000 deaths respectively), the Observatório Nacional de Saúde (ONSA) - Instituto Nacional de Saúde Dr. Ricardo Jorge, together with the Vigilância Previsão e Informação - Instituto de Meteorologia, created a surveillance system called ÍCARO, which has been in operation since 1999. ÍCARO identifies heat waves with potential influence on mortality [1].
Before the end of the 2003 heat waves, ONSA had produced a preliminary estimate of its effect on mortality. The results based on daily number of deaths from 1 June to 12 August 2003 were presented within 4 working days. Data was gathered from 31 National Civil reg...
The Environment Agency of England and Wales obtained European Community Initiative INTERREG IIIB funding for a project called ICReW—Improving Coastal and Recreational Waters. The project consists of seven pilot actions aiming to contribute to the reduction of pollution, to enhance water quality, and to ensure that land-use practices and recreational activities
can exist side by side without impacting public health. One of these actions is to identify and develop a common methodology
for source-tracking fecal pollution, for regulatory purposes, over a wide geographical area in Europe. In order to do this the Department for Environment, Food and Rural Affairs, UK, sponsored the first international workshop on the subject. Key
researchers from around the world were invited to attend to recommend the most appropriate method(s) for devel...
Hereditary haemochromatosis (HH) is an autosomal recessive disorder
characterized by excessive intestinal iron absorption resulting in increased
pathological body iron stores. It is typically associated with homozygosity
for the c.845G>A (p.C282Y) mutation in the HFE gene. However, other
HFE alterations have been reported in affected individuals but their association
with the disease is unclear. This study analysed the functional consequences
of two HFE mutations, c.829G>A (p.E277K) and c.884T>C(p.V295A). Firstly, it was shown that c.829G>A affects the HFE splicing by
diminishing the full length HFE and ivs4_66bp inclusion transcript levels,
while increasing the amount of exon 4 skipping transcript. Immunofluorescent
techniques showed that the HFE_E277K protein had a diffuse distribution(similar to HFE_C282Y) while HFE_V295A p...
Nonsense-mediated decay (NMD) is an mRNA surveillance pathway that selectively recognizes and degrades defective
mRNAs carrying premature translation-termination codons. However, several studies have shown that NMD also targets
physiological transcripts that encode full-length proteins, modulating their expression. Indeed, some features of
physiological mRNAs can render them NMD-sensitive. Human HFE is a MHC class I protein mainly expressed in the liver that,
when mutated, can cause hereditary hemochromatosis, a common genetic disorder of iron metabolism. The HFE gene
structure comprises seven exons; although the sixth exon is 1056 base pairs (bp) long, only the first 41 bp encode for amino
acids. Thus, the remaining downstream 1015 bp sequence corresponds to the HFE 39 untranslated region (UTR), along with
exon seven. Therefore...
